Once considered mainly 'brain glue,' astrocytes' power revealed

A type of cell plentiful in the brain, long considered mainly the stuff that holds the brain together and oft-overlooked by scientists more interested in flashier cells known as neurons, wields more power in the brain than has been realized, according to new research published today in Science Signaling.

Neuroscientists at the University of Rochester Medical Center report that astrocytes are crucial for creating the proper environment for our brains to work. The team found that the cells play a key role in reducing or stopping the electrical signals that are considered brain activity, playing an active role in determining when cells called neurons fire and when they don't.

That is a big step forward from what scientists have long considered the role of astrocytes ? to nurture neurons and keep them healthy.

"Astrocytes have long been called housekeeping cells ? tending to neurons, nurturing them, and cleaning up after them," said Maiken Nedergaard, M.D., D.M.Sc., professor of Neurosurgery and leader of the study. "It turns out that they can influence the actions of neurons in ways that have not been realized."

Proper brain function relies on billions of electrical signals ? tiny molecular explosions, really ? happening remarkably in sync. Recalling the face of a loved one, swinging a baseball bat, walking down the street ? all those actions rely on electrical signals passed instantly along our nerves like a molecular hot potato from one brain cell to another.

For that to happen, the molecular brew of chemicals like sodium, calcium and potassium that brain cells reside in must be just right ? and astrocytes help to maintain that balanced environment. For instance, when a neuron sends an impulse, or fires, levels of potassium surrounding the cell jump dramatically, and those levels must come down immediately for the brain to work properly. Scientists have long known that that's a job for astrocytes ? sopping up excess potassium, ending the nerve pulse, and restoring the cells so they can fire again immediately.

In the paper in Science Signaling, Nedergaard's team discovered an expanded role for astrocytes. The team learned that in addition to simply absorbing excess potassium, astrocytes themselves can cause potassium levels around the neuron to drop, putting neuronal signaling to a stop.

"Far from only playing a passive role, astrocytes can initiate the uptake of potassium in a way that affects neuronal activity," said Nedergaard. "It's a simple, yet powerful mechanism for astrocytes to rapidly modulate neuronal activity."

Nedergaard has investigated the secret lives of astrocytes for more than two decades. She has shown how the cells communicate using calcium to signal. Nearly 20 years ago in a paper in Science, she pioneered the idea that glial cells like astrocytes communicate with neurons and affect them. Since then, has been a lot of speculation by other scientists that chemicals call gliotransmitters, such as glutamate and ATP, are key to this process.

In contrast, in the latest research Nedergaard's team found that another signaling system involving potassium is at work. By sucking up potassium, astrocytes quell the firing of neurons, increasing what scientists call "synaptic fidelity." Important brain signals are crisper and clearer because there is less unwanted activity or "chatter" among neurons that should not be firing. Such errant neuronal activity is linked to a plethora of disorders, including epilepsy, schizophrenia, and attention-deficit disorder.

"This gives us a new target for a disease like epilepsy, where signaling among brain cells is not as controlled as it should be," said Nedergaard, whose team is based in the Division of Glia Disease and Therapeutics of the Center for Translational Neuromedicine of the Department of Neurosurgery.

The first authors of the paper are Fushun Wang, Ph.D., research assistant professor of Neurosurgery; and graduate student Nathan Anthony Smith. They did much of the work by using a sophisticated laser-based system to monitor the activity of astrocytes in the living brain of rats and mice. The work by Smith, a graduate student in the University's neuroscience program, was supported by a Kirschstein National Research Service Award from the National Institute of Neurological Disorders and Stroke (NINDS).

Other authors from Rochester include Takumi Fujita, Ph.D., post-doctoral associate; Takahiro Takano, Ph.D., assistant professor; Qiwu Xu, technical associate; and Lane Bekar, Ph.D., formerly research assistant professor, now at the University of Saskatchewan. Also contributing were Akemichi Baba of Hyogo University of Health Sciences in Japan, and Toshio Matsuda of Osaka University in Japan.

Nedergaard notes that the complexity and size of our astrocytes is one of few characteristics that differentiate our brains from rodents. Our astrocytes are bigger, faster, and much more complex in both structure and function. She has found that astrocytes contribute to conditions like stroke, Alzheimer's, epilepsy, and spinal cord injury.

"Astrocytes are integral to the most sophisticated brain processes," she added.

The latest paper caps a remarkable run for the Nedergaard group. In two months the team has published a paper on adenosine in the Proceedings of the National Academy of Sciences, a paper on spinal cord injury in the Journal of Neuroscience, and two papers in Science Signaling on communication between neurons and astrocytes.

Love Is...Some Ground Up Hamburg

Ladies, I'm going to brag on my husband today. I believe he deserves it.


The other night, we had family taco night, which we hadn't done in a while. My hubby came home from a long day at work and went to directly making dinner while I watched our daughter play in the living room.


He cooked up the hamburger for our tacos into two separate dishes; one with the taco seasoning and one without just for me because he knows I don't like the spiciness of the seasoning. Then he diced up the tomatoes for me (even though he doesn't like them himself). And spread out all the fixings that my son and I like on the table.






We took turns eating so that one of us could feed our toddler and the other could enjoy dinner while it was still warm.


When dinner was finished, I mentioned I wanted to use the extra hamburger for my lunch the next day. He apologized that there wasn't a lot of meat left since he wasn't sure how much to leave out for me that wasn't seasoned. I was a little disappointed that there was just a little bit left, and I guess he could see that in my face (though I told him it was fine, I could make do with what we had).


He went out to the pharmacy to pick up some things that I needed and came home. Then I saw him in the kitchen again cooking and asked him what he was making, thinking that maybe he was making his lunch for work ahead of time.


He told me that he was cooking hamburger.


He had stopped by the grocery store on the way home and picked up some more so that I could have it for my lunch. And he took the time to cook it up for me and store it in the fridge to save me from having to do it.


When I put together my lunch the next day, I couldn't help but smile and be reminded of the sweet thing that he did for me. It was something simple, but that simple thing was done with so much love behind it.








I am one blessed woman to be married to a man who demonstrates love every day with simple actions.

NEW YORK (AP) ? A very tiny puppy is sitting inside a coffee mug, trying without success to escape by scratching its little paws against the slippery surface. In recent days, this famous dog has been placed alongside a variety of small objects ? an iPhone, a business card and a tape measure, among other things ? to demonstrate just how tiny she really is.

This is Beyonce, a female dachshund mix who was born March 8 to a rescue dog that was found abandoned, wandering the streets of San Bernardino, Calif. At birth, she weighed just 1 ounce and could fit into a teaspoon. Her caretakers say she's one of the smallest puppies ever born full-term ? and her story of unlikely survival has captured the attention of people all over the world.

"We had an ultrasound done, and they actually saw that there were five puppies, but one was probably going to be stillborn," says Beth Decaprio, executive director of the Grace Foundation of Northern California, which rehabilitates abused and neglected animals. "It didn't look like a viable baby."

Beyonce wasn't breathing when she was born at the foundation's farm in El Dorado Hills, Calif. A veterinarian tried to revive her by performing chest compressions. Then he passed her over to Decaprio.

"I blew a couple little breaths in her mouth," Decaprio says. "And she started to breathe on her own."

At 3 weeks old, Beyonce is now about the size of an iPhone, but she isn't cowed by her much bigger brothers and sisters. She crawled on top of the wriggling puppies as they nursed, trying to push them out of the way.

"They just consider her another sibling," Decaprio says. "But she is a survivor, and she's a tough little one."

Beyonce, whose caretakers wanted to give her a "big" name, is not yet up for adoption, though the foundation has received hundreds of requests from people who want to take her home. When she's stronger ? and a bit bigger ? the foundation will decide whether to give her away.

The shelter has taken the puppy on a media tour, including big media stages in New York.

Decaprio hopes the little puppy will inspire people to adopt shelter dogs and save them from being destroyed. Many shelters keep rescue dogs for only 12 hours before they must be put down, she says.

"What's most important about the story is that people are aware of the tiny, little, wonderful miracles that come out of the shelter that sadly are euthanized each and every day," she says. "Beyonce and her family are lucky, but we know that most of these don't get that kind of chance."

SolarCity Charges Into Home Efficiency With Admirals Bank ...

The solar energy industry was still bemoaning the high upfront costs of rooftop solar installations when SolarCity helped pioneer the solar lease, which has virtually eliminated that hurdle. Now, with financial backing from Admirals Bank, SolarCity has set out to do the same thing for the home energy efficiency upgrade business.

?By making energy-saving measures more accessible and affordable,? said SolarCity CEO Lyndon Rive, ?we?re doing for energy efficiency what we?ve done for solar power.?

Taking advantage of the new Boston-based Admirals Bank backing, SolarCity is expanding its energy efficiency service, which it began offering in addition to rooftop solar installations in 2010, to customers in Connecticut, Maryland, Massachusetts, New Jersey, New York and Washington, D.C. The company already has 5,000-plus efficiency projects completed or underway in Arizona, California, Colorado, Oregon and Texas.

?We?re trying to bring our customers a full range of clean, affordable alternatives to utility power,? explained SolarCity spokesperson Jonathan Bass. Electricity generated by fossil fuels for a single home, a federal study found, puts more carbon dioxide into the air than two average cars.

Admirals Bank, a federal savings bank with a history as a home improvement lender, will make available between $2,500 and $15,000 per customer, Bass said. That should readily meet the cost of the typical upgrade which, according to SolarCity Manager of Energy Efficiency Services Levi Blankenship, is between $4,000 and $12,000. That investment, Blankenship added, typically reduces a homeowner?s combined natural gas and electricity costs by 20 percent to 50 percent.

A Department of Energy (DOE) study found that the U.S. spends $241 million yearly on home energy bills. That?s one in five of the dollars the U.S. spends on energy. It is an average of $1,900 per family.

?We saw with solar that if we reduce or eliminate the upfront costs,? Bass explained, ?it makes it available to a new set of customers.? The company?s expectation is that the same thing will happen with energy efficiency.

The SolarCity process begins, Blankenship said, with an on-site home energy evaluation that diagnoses the cause of high utility bills and energy losses. Proprietary SolarCity software does four-billion-plus calculations for each home evaluation and produces a report across nine efficiency categories. SolarCity uses the report to prioritize recommended home improvement options based on energy and cost savings.

?Typically,? Blankenship said. ?There two categories of improvements.? Over half a building?s energy consumption is for ?space conditioning,? he explained, ?either heating or cooling. So the building shell, or envelope, is a huge component.?

A SolarCity 250-home analysis, incorporating DOE statistics, found that about $40 of every $100 spent on heating and cooling is lost to duct and air leakage. In their evaluation, SolarCity people identify the amount of ?re-insulation? and ?air sealing? needed ?to retain heat in the winter and keep it out in the summer so that the furnace and air conditioner do not need to run as often.?

Once they are sure the building shell is efficient, Blankenship said, ?We turn our attention to the mechanical equipment itself. The furnace, the air conditioner, the heat pump, the water heater, the equipment that provides conditioned air. We want to be sure it is efficient and sized for the house.?

It is ?the granularity of the software evaluation,? Blankenship added, ?that makes it possible for us to eliminate client and confusion concerns that may have slowed adoption in the past. The software, coupled with the loans that will make it possible to eliminate upfront costs, is really exciting.?

SolarCity?s efficiency financing already supports a range of local energy efficiency loan and rebate programs. The Admirals Bank-backed Home Energy Loan financing option is expected to further reduce and/or eliminate the upfront cost of energy efficiency upgrades.

?We will work with programs that are out there in our service territories and direct our customers to the best option for them,? Bass said. ?But in places where there is no efficiency program, we wanted to give customers the option of evaluations and upgrades.?

SolarCity and Admirals Bank intend to make the loan process as convenient and accessible for customers as possible. Eligible customers will be able to choose between a one-year ?Save Now, Pay Later? option and 3-year or 10-year ?Pay As You Go? options. The one-year option can be interest-free.

?Commonly, there are utility, county, and/or state rebate programs, but the rebates don?t get paid immediately. There can be a several month lag.? The one-year option gives homeowners time to collect and use the rebates and pay off the balance without incurring interest charges. ?If the loan is repaid within one year,? Bass explained, ?there is no interest.?

?And they start saving energy and money right way,? Blankenship added.

(AP) ? Alisha Carter-Camp had a new job, a wedding to plan and a 26th birthday to celebrate with a family cookout and toasts to the birthday girl in a yard full of children. By the end of the night, she was among eight dead, including six children, in one of this West Virginia city's deadliest house fires in decades.

The blaze tore through the two-story home while the family slept early Saturday, hours after the last guest had left Carter-Camp's party, authorities said. The dead children ranged from 18 months to 8. A seventh child, a 7-year-old boy, was hospitalized on life support.

The cause was under investigation, although arson wasn't suspected, Charleston Mayor Danny Jones said. The fire started about 3:30 a.m. on the first floor. Jones said the home had just one working smoke detector; the city requires several. A building inspection that had been scheduled for last month didn't happen because only children were home at the time.

A children's picnic table, chairs and an umbrella were overturned in the yard of the home, roped off by police tape on a corner in a neighborhood tightly packed with small houses in north Charleston. Flames and smoke blackened the front of the house Two huge front windows were shattered, and what appeared to be an opening for an upstairs air conditioner was stuffed shut with clothes.

Alisha Carter-Camp, who would have been 26 Saturday, was among those killed, Jones said. She had been working as a hotel clerk for six months and told neighbors she planned to get married in June and move to Pittsburgh.

Four of the victims were 3 years old: Jeremiah Camp, Elijah Scott and two children who were only identified by their first names. Also killed were Keahna Camp, 8, Emanuel Jones, 18 months, and an adult, Alex Seal.

Bryan Timothy Camp, 7, was hospitalized in critical condition. One adult survivor, Latasha Jones Isabell, went to the hospital but it wasn't clear if she was treated. Authorities said all the victims were related, but they weren't sure how and didn't know how many lived full-time in the home.

"We have reason to believe that a lot of the people stayed in the house more than one night and maybe on a weekly basis," Jones said. "These people may have had residences in other places, but a lot of people lived in this house."

Carter-Camp and her two children were staying with her sister at the home, said Roxie Means, who lives down the street.

The party started Friday afternoon with a cookout and toasts of wine.

"They were nice people drinking a glass of wine," Roxie Means said. "They weren't drunk. They weren't overdoing anything."

Roxie's daughter, 14-year-old Cassie Means, said she noticed lit candles inside the home when she attended the party Friday night.

Roxie Means said Isabell, 24, was smoking a cigarette outside, noticed the fire and came running to Means' home in the middle of the night and started "beating down the door."

The home was engulfed in flames when firefighters arrived. When they went inside, they immediately came across five victims and "started realizing there were a lot of people in this house, a lot of children."

Jones said he was devastated by the news of the fire ? the deadliest in the state capital since seven firefighters perished while battling a fire at a Woolworth department store in 1949.

"I was with my children and I just grabbed them and hugged them, because I have a 5-year-old and a 4-year-old," he said. "I walked up there and caught a glimpse of some fatalities and it's something that's hard to grasp. The fact that there are (six) dead children, it's unimaginable."

Rusty Eaton, general manager at the Holiday Inn Express Charleston Civic Center, said Carter-Camp was one of his employees. Eaton said he was notified of her death by her mother Saturday morning.

"Everybody's taking it pretty tough, including myself," Eaton said. "It's a tough thing. It's something you hope you never have to deal with."

He said Carter-Camp had worked at the hotel's front desk for about six months and also helped audit financial paperwork at night. He said that she felt at ease speaking with anyone and that "you didn't have to teach (her) personality."

"She had one of those infectious smiles, never met a stranger. She had personality to spare," Eaton said. "She was great with our customers, great with her co-workers. Certainly, she'll be sorely missed by us."

By midmorning, police had pulled the bodies out. On Saturday afternoon, police searched Carter-Camp's vehicle still parked outside.

"She worked seven days a week, had her kids here," Roxie Means said. "I love the kids. That's really what hurts us."

Jones said the home had one working smoke detector on the bottom floor, but it was on a counter and wasn't properly mounted to a ceiling. Another was found in a basement area but didn't work. The city requires smoke detectors in basements, one in every bedroom and in hallways.

A city building inspector had made arrangements with the owner, Deloris Shamblen, to inspect the home on Feb. 28. Carter-Camp had signed a permission slip for an inspection of the property, but when the inspector arrived, a child answered the door and asked the inspector to come back when an adult was there, Jones said.

"Had we seen the fact that they did not have the proper number of smoke detectors, we might have saved a lot of lives," Jones said.

A telephone message left with Shamblen wasn't immediately returned Saturday.

Roxie and Cassie Means stood outside the burnt-out house and remembered the kids playing in front of it just hours earlier. Cassie Means said she had made the kids a promise; they wanted to know if she would be back on Saturday.

"'Cassie, are you coming over to play with us tomorrow?' I said, 'yeah.'" she recalled.

The child continued, "'You promise me you'll be here tomorrow?'" Cassie Means recalled. "I said, 'I promise you I'll be here when you wake up to play with you. I'll be here right when you wake up."

How an English pub paid the price for failing to choose

Here?s an example of what can happen, if you try and attract both ends of the market.

Around 5 years ago, a friend asked me if I would speak with his sister, who owned a village pub and restaurant in Kent, England. He said she needed help and he was right. The problem she had was immediately obvious. She was trying to position the pub simultaneously as a place that sold cheap booze AND as a family venue with great food.

Here?s what happened. The cheap booze attracted people who were, by her own admission, young, loud and prone to bad language. This stopped most families from visiting the place more than once, as most people don?t want their kids around that kind of atmosphere. Equally, it wasn?t the kind of atmosphere you would choose if you wanted to go out for a nice meal either. So, a very small percentage of diners ever returned.

There wasn?t enough profit from the cheap booze crowd to sustain the business, however, because the boozers were stopping the highly profitable diners from eating there, she said she didn?t want to lose the cheap booze income, for fear of having nothing. So, she ended up wasting thousands on advertising and promotions aimed at diners and families, which did attract some people, but very few returned. After 18 months in business, she ran out of money and then she ran out of places to borrow money.

Interestingly, the pub was then bought by someone who transformed it into a very successful country restaurant, bar and seminar / meeting venue, with a reputation for great food and a warm, welcoming atmosphere. The place has a clear identity now, as a quality venue. It attracts people who are happy to pay a premium for quality. It doesn?t attract people looking for a cheap place to get drunk.

We have to choose

I wrote recently about how we need to match our fees or prices to our promises. Well, we also need to check that our business is balanced correctly, that people know?who we are and where we fit into the quality / price spectrum. For example, a designer with promotions all over their website for bargain $50 logo designs, is going to have trouble convincing us they are worth paying $5,000 for their premium design service. We know they do $50 work, that they work at the bottom end of the scale - The best designers don?t do that.

Where do you pitch your prices or fees and what messages do you think that gives people? Are you trying to attract both, premium clients and budget conscious clients? If so, are you aware this usually makes you look like poor value, for the premium clients?

Marketing Tip

Take some time to review the type of client you are attracting (premium focused or budget conscious.) If you want more premium clients, but tend to attract the fee sensitive crowd, there?s ALWAYS a reason. Start by looking at your marketing messages and pay special attention to anything that suggests you serve the cheap end of the market.

Just like the pub owner I mentioned earlier, we have a decision to make, if we want to attract the best clients and the most attractive fees.

House GOP unveils budget blueprint

WASHINGTON (AP) ? Conservative Republicans controlling the House unveiled a budget blueprint Tuesday that combines slashing cuts to safety net programs for the poor with sharply lower tax rates in an election-year manifesto painting clear campaign differences with President Barack Obama.

The GOP plan released by House Budget Committee Chairman Paul Ryan would, if enacted into law, wrestle the deficit to a manageable size in short order, but only by cutting Medicaid, food stamps, Pell Grants and a host of other programs that Obama has promised to protect.

To deal with the influx of retiring Baby Boomers, the GOP budget reprises a controversial approach to overhauling Medicare that would switch the program ? for those under 55 today ? from a traditional "fee for service" framework in which the government pays doctor and hospital bills to a voucherlike "premium support" approach in which the government subsidizes purchases of health insurance.

Republicans say the new approach forces competition upon a wasteful health care system, lowering cost increases and giving senior more options. But Democratic opponents of the idea say the new system ? designed by Ryan and liberal Sen. Ron Wyden of Oregon ? cuts costs too steeply and would provide the elderly with a steadily shrinking menu of options and higher out-of-pocket costs.

This year's GOP measure would produce deficit estimates that are significantly lower than a comparable measure passed by the House a year ago, claiming deficit cuts totaling $3.3 trillion ? spending cuts of $5.3 trillion tempered by $2 trillion in lower taxes ? below Obama over the coming decade. The deficit in 2015, for example, would drop to about $300 billion from $1.2 trillion for the current budget year. Last year's GOP draft called for a 2015 deficit more than $100 billion higher.

The measure would cut spending from $3.6 trillion this year to the $3.5 trillion range in 2013 and freeze it at that level for two more years.

The GOP plan doesn't have a chance of passing into law this year but stands in sharp contrast to the budget released by Obama last month, which relied on tax increases on the wealthy but mostly left alone key benefit programs like Medicare.

The resulting political battle is sure to spill beyond the Capital Beltway into the presidential race and contests for control of the House and Senate this fall. As if to underscore that reality, Ryan released a campaign-style video Monday evening telling viewers that "Americans have a choice to make" in a none-too-subtle appeal to voters.

"It's up to the people to demand from their government a better budget, a better plan, and a choice between two futures," Ryan said. "The question is: which future will we choose?"

The Budget panel is slated to debate and vote on the measure Wednesday and in hopes of a vote by the full House next week.

The Senate has no plans to debate a budget and will instead rely on last summer's bipartisan budget and debt pact to govern this year's round of spending bills.

The annual budget debate in Congress plays out on an arcane battlefield of numbers and assumptions, often difficult to understand even by Capitol Hill veterans. Basically, however, the so-called budget resolution sets broad parameters for follow-up legislation. Sometimes that is just a round of agency budget bills; other times lawmakers take on taxes and benefit programs like Medicare whose budgets otherwise run on autopilot.

The lower deficit figures build on cuts to annual agency budgets imposed last year and rely on new savings comes from benefit programs outside Social Security and the costly Medicare and Medicaid health care programs for the elderly and the poor. That means big cuts to food stamps, student loans, welfare, farm subsidies and other programs whose budgets now mostly run on autopilot.

On taxes, the measure calls for eliminating a host of tax deductions and credits in order to produce a far simpler income tax code with just two rates for individuals: 10 percent and 25 percent. But Ryan doesn't say the income levels at which the new rates would apply, nor does he specify which popular tax breaks ? like the child tax credit or the mortgage interest deduction ? might be spared.

Medicaid would be sharply cut and awarded to states as a flexible block grant.

Just as Obama's budget was dead on arrival last month with Capitol Hill Republicans, the House GOP plan is a nonstarter with Democrats controlling the Senate.

On Monday, two powerful Senate committee chairmen sent top House GOP leaders a letter protesting a GOP plan to cut agency operating budgets funded annually by Congress below levels negotiated just last summer. Instead of going with a $1.047 trillion cap on agency budgets as called for under last summer's debt and budget pact, the House panel is looking at cutting domestic agencies by $19 billion more.

Senate Budget Committee Chairman Kent Conrad, D-N.D., and Appropriations Committee Chairman Daniel Inouye, D-Hawaii, warned that breaking with the agreement only guarantees delays later this year and "represents a breach of faith that will make it more difficult to negotiate future agreements."

Also at issue, though, are across-the-board spending cuts set to take effect in January, punishment for the failure of last year's supercommittee to come up with a new package of $1.2 trillion in deficit cuts over the next decade as part of last summer's deal to let the government keep borrowing.

Those cuts, including $55 billion from defense accounts and $43 billion from non-defense accounts approved by lawmakers each year, are universally opposed by defense hawks and liberals alike.

The GOP plan would reverse the cuts by requiring various committees and try come up with at least $261 billion in other savings over the coming decade, including curbs to food stamps, federal employee pensions, and further cuts to federal health care programs. Republicans are likely to reprise a bid to tighten oversight of the child tax credit to make sure illegal immigrants don't claim it.

The measure would produce a $797 billion deficit in the upcoming 2013 fiscal year, as opposed to $977 billion under Obama's budget. The deficit would fall to $241 billion by 2016, compared to a $529 billion deficit in 2016 under Obama's plan.

"The country wants to be spoken to like adults, not pandered to like children," Ryan said Tuesday on "CBS This Morning."

The Wisconsin Republican said, "If you want to save Medicare and keep it from going bankrupt, you must reform the program, and that's what we intend to do."

Ryan said he has grown weary of the GOP being accused of endangering the benefits that senior citizens have come to expect. "We preserve the program for people in and near retirement," he said. "We want to take all the empty promises our government is making and make sure they're not broken promises."
